Can't remember exactly, but it was a morris tune.
When I joined my first morris side we used to do lots of fetes, and while everybody else was off buying cakes and plants I used to stay behind and guard all the belongings, which included a melodeon or two.  I amused myself by picking out tunes, and that's how I started to play the box.
The treble end was easy....the bass end wasn't!

I think it was Shepherd's Hey and I used my cassette recorder to record bass and treble separately then play the other side  to get the idea.  It was like rubbing head and belly at same time

First "real" tune was Egan's Polka straight out of the Mallinson book. Still one of my favorites

Mine was possibly Constant Billy, or some other morris tune, mayne Highland Mary.

I think the first tune I cracked properly with both treble and basses was Bluebells of Scotland, then Princess Royal I think. I still love the latter, but cannot abide the former any more. The word boring (both tune and dance) doesn't even begin to cover it.

Winster Gallop..  as it was the first tune in 'Handbook For Melodeon'

Constant Billy is on page two.. that must have been the 2nd one

Mine was Astley's Ride, closely followed by Star of the County Down, which is partially because they're two of my favourite tunes to morris dance to!

After my decade-long love affair with a 30 string harp, I was naturally immersed in O'Carolan tunes. My first melodeon tune was Fanny Power. I still love all of those songs and most fit nicely with melodeon or concertina.

First tune on a BC black dot was An Coinniachaen (aka Fairy Lullaby). I can even play it with both hands. My nine year old daughter is playing it better than me now (her first tune too).

Google google. Gosh, I've played that one for years without ever knowing its name. It appears Ireland didn't have that book either. I cannot imagine that Carol an(n) was a 60's feminist?  ;D

Yup - I could even name that one! Like both of 'em - perhaps more 'chords' in them than your usual Irish gig/reel (just speculating)
