Th first test of whether an accordion in tune is whether it sounds good to you the player (and possibly the people you play with). If it sounds sour, or there is just something you can't quite identify but it doesn't seem quite right, then it's time to have it tuned. You can narrow that down a bit by comparing the sound of the same note in different rows, and if you can use stops to get a single reed sound then try playing octaves and listen for beats.  If you have a LM voice available there should be no beats on any single note.
